Tβ4 has been used at our clinic with a great deal of success in patients suffering from Lyme disease or other infectious and/or inflammatory conditions. Tβ4 is very well tolerated and has not been found to cause any significant side effects. It can be taken on its own or in conjunction with an existing therapy, making it a versatile and valuable drug.
The weight lost by the 1mg group was slightly more than that achieved by the world’s largest selling prescription obesity medication in similar trials over the same period, without its troublesome side effects. Injections of other compounds along with IGF-1 (which is a popular practice) can also cause serious health issues. The idea is that after an user administers a GHRP (like Ipamorelin) along with IGF-1, a selective pulse is then sent that stimulates the hypothalamus and pituitary to release even more growth hormone. But this may result in an eventual negative feedback loop that leaves you unable to produce your own growth hormone and stuck on injections forever. GHRP and synthetic HGH use has also been shown to cause joint pain, huge spikes in cortisol, excessive hunger, and splitting headaches.

Other peptides amplify the body’s response to its own testosterone site-specifically within muscle and bone only. Steroids, on the other hand, produce their highly desired anabolic effects by acting on these receptors, but also come coupled with the less-sought-after side effects of acne, oily skin, hair loss, aggression, breast enlargement and testicular shrinkage. It is for this reason, these amino acid chains are serving as an increasingly popular alternative to steroids amongst aesthetic gym goers, providing many of the anabolic effects without the unwanted side effect profile.
One proposed mechanism for reduced natriuretic peptide concentrations in obesity is the relative abundance of natriuretic peptide clearance receptors (NPR‐C) in adipose tissue.13, 22 Elevated insulin has also been linked to increased expression of NPR‐C in obese subjects.23 On the other hand, plasma Nt‐proANP and Nt‐proBNP levels are reduced in obesity to a comparable degree as the mature peptides. Because the pro‐peptides are not known to bind to NPR‐C, impaired synthesis or secretion likely plays a role in obesity.
Other studies have had similar results with regards to the effect of collagen supplementation in helping to promote fullness. One study consisted of 24 healthy adults testing the satiety effects of various protein supplements. The subjects had two breakfast meals with specific types of protein in each: one meal had alpha-lactalbumin, gelatin, or gelatin + tryptophan (TRP) (Breakfast 1) and the other meal contained casein, soy, whey, or whey + glycomacropeptide (GMP) (Breakfast 2). The study found that Breakfast 1, which included gelatin (collagen), was 40% more satiating than Breakfast 2, which did not contain gelatin or collagen. Additionally, the participants who ate Breakfast 1 with gelatin ended up consuming less calories for lunch. Researchers concluded that gelatin increases satiety, which can lead to subsequent reduced energy intake, thereby promoting weight loss (2).
Cerebrolysin: A mixture of peptides extracted from pig brain that supports the development and function of nerve cells, cerebrolysin can be used to treat Alzheimer’s disease and vascular dementia but because the drug needs to be injected daily, it doesn’t have a clear benefit over existing oral medication. It’s not known to have any major side effects. In sports, cerebrolysin used to enhance mental function and “defog” the brain, although there is no good evidence that it has any effect in healthy individuals. It’s not currently listed as banned under WADA, either directly or via similarity of mechanism to banned peptides.
Nevertheless, the hypothesis that AOD9604 does not activate the hGH/IGF-1 axis had to be tested in humans. The studies presented here confirm the in-vitro results. In these studies no clinically relevant changes of IGF-1 levels were observed and no differences to the placebo treatment were found. Together with the lack of any other symptoms associated with known IGF-1 mediated effects, such as sodium retention, tissue oedema, hypertension, or impaired glucose tolerance, the results demonstrate that AOD9604 does not activate the hGH/IGF-1 pathway and therefore has no growth promoting effect.

There is much confusion about the significance of the banned peptide at the centre of the AFL doping scandal - and perhaps that is not so surprising. The company behind AOD-9604 knew back in 2007, after six clinical trials of 925 patients, that the drug had completely failed to treat obesity. But after burning through some $50 million developing the drug, Metabolic Pharmaceuticals Pty Ltd, which owns the patent, has continued to spruik its so-called body-enhancing benefits.
In vitro and in vivo investigations revealed a specific region within the hormone molecule that is responsible for the molecular events associated with lipid metabolism [18, 24, 25]. AOD9604 is a peptide fragment of the C-terminus or lipolytic domain of hGH (hGH177-191), with an additional tyrosine residue at the N-terminal end for stabilization. In vitro and in vivo experiments have shown similar effects of AOD9604 and hGH on lipid metabolism when chronically applied to mice [20, 21]. Interestingly, AOD9604 mimics the effect of hGH on lipid metabolism, without having growth promoting or pro-diabetic effects. The safety and tolerability of AOD9604 has been studied in the human clinical trials described in this paper.

Following amplification, PCR products were electrophoresed on 1.3% agarose gels and transferred onto Hybond N+ membranes (RPN 303B, Amersham Pharmacia Biotech) by Southern blotting in 0.4 M NaOH/1 M NaCl. The β3-AR product bands were normalized against the β-actin control, averaged, and RNA isolated from treated animals was expressed against control animals.

Ipamorelin, like other growth hormone peptides, is used to increase production of your own growth hormone levels. Technically, it is a “protein-peptide hormone” which means that it consists of 70 amino acids bonded together. Just like the peptides I’ve written about in the past, this means that it must be injected, because otherwise IGF-1 simply degrades in the gut, rendering it useless. Your own human growth hormone release promotes the synthesis of IGF-1 in your liver (and to smaller amounts, synthesis of IGF-1 by your muscles), your liver and muscles then synthesize IGF-1 and then, in the case of your liver, subsequently package the IGF-1 with binding proteins for transport into the blood. In a type of anabolic positive-feedback loop, IGF-1 then further increases growth hormone’s anabolic effects.
In ob/ob mice, both AOD9604 and hGH reduced both white and brown adipose tissue mass and increased β3-AR RNA expression. This suggested that an elevation inβ 3-AR RNA expression is associated with increased fat metabolism and a reduction in the fat tissue mass in the ob/ob mouse model. Obese mice have lower levels ofβ 3-AR expression in their adipose tissues than lean mice, shown in this study and others (14). The ability of AOD9604 and hGH to increase the level ofβ 3-AR RNA expression in obese mice to a level that is comparable to those in lean mice is an exciting finding. However, it must also be considered that both hGH and AOD9604 may influence the expression of other members of the adrenergic pathway, such as the β1-ARs, hormonesensitive lipase, and signaling proteins, which are all expressed in adipose tissue and associated with lipolysis. The importance of the change inβ 3-AR expression with AOD9604 and hGH in humans is not established and will depend on the use of potent and selectiveβ 3-AR agonists that are active at the human receptor.

Since actual HGH shuts down the body's natural pituitary gland, when you stop injecting it, your body's own ability to produce Growth Hormone is hindered and you will suffer a rebound of negative side effects such as fat gain, muscle loss and loss of skin tone/elasticity. Growth Hormone (GH) exhibits its muscle building effects mainly after its conversion to IGF-1 (Insulin-Like-Growth Factor). Hey Elvia, do you think that using DAC for short periods of time at your proposed dose is still effective for fat loss? Say for four weeks at 4mg a week? During the hydrolysis process, whole proteins are broken down into smaller peptide fragments. These can sometimes be as short as two or three amino acids in length (known as di- and tri-peptides). The benefits of hydrolysed protein have been outlined in detail elsewhere. In short, hydrolysed proteins are much faster absorbed than other forms of protein. It has been shown that hydrolysed whey protein can increase the time of recovery compared to whey protein isolate (WPI) (Buckley et al, 2010).

In this study, plasma glycerol was also assayed as a measure of lipolytic rate. As shown in Fig. 5C, both AOD9604 and hGH increased glycerol levels following treatment in the WT mice, indicative of enhanced lipolysis. In the β3-KO mouse, however, no effect was observed with AOD9604. A significant increase in plasma glycerol from controls was observed following hGH treatment, but this was markedly less than that observed in the WT mouse. These data indicate the importance of β3-AR in the lipolytic response to AOD9604 in these animals and the necessity of the receptor for the chronic effectiveness of AOD9604 and hGH on fat reduction.

These results also demonstrate that, unlike hGH, AOD9604 has no negative effect on carbohydrate metabolism. This was previously demonstrated in mice. Chronic administration of hGH to ob/ob mice depressed glucose oxidation and increase plasma glucose levels. AOD9604 had no such effects, no changes in circulating plasma glucose in either lean nor obese ob/ob mice have been observed [20]. AOD9604 did not cause hyperglycemia or affect insulin sensitivity in rats and mice [18, 20].

AOD9604 has been scientifically proven safe and side-effect free (2), and is apparently very difficult to detect in the blood.
Obesity affects as many as one in five adults in developed countries. Metabolic estimates that the potential worldwide market for effective obesity drugs is as much as $20 billion, far more than the current market of $1.5 billion. This is partly due to the potential need for chronically obese people to take obesity drugs such as AOD9604 and also because patients are put off using currently available drugs because of their marginal efficacy or side-effects.
Similar to GHRP 2, this peptide is a more potent releaser of growth hormone, also acting on the ghrelin receptors of the anterior pituitary. Also like GHRP 2, GHRP 6 leads to increased growth hormone production, increased lead body mass, and decreased adiposity. Due to the peptide’s ghrelin-like properties, administration can lead to increased appetite.
