The knee joints were dissected after euthanasia. The medial and lateral condyles of the femur and tibia were examined for gross morphological changes. The degree of cartilage degeneration on the lateral part of the femoral condyle was assessed using the scoring system devised by Yoshimi et al. [12]. This area was assessed because the intra-articular collagenase injections had caused most damage at this area [13]. Normal cartilage was scored as 0, softened cartilage as 1, fibrillated cartilage as 2, eroded cartilage as 3, ulcerated cartilage as 4, and loss of cartilage as 5.

AOD9604 is a new synthetic peptide fragment that comprises a modified 15 amino acid region of GH with a tyrosine component to help stabilize the molecule. Similar to GH, AOD9604 aids weight reduction in rodent models of obesity and was originally developed for the treatment of obesity in humans [11]. Additionally, it does not stimulate the production of IGF-1 [10], has positive effects on the differentiation of adipose mesenchymal stem cells into bone, and was found to promote proteoglycan and collagen production in isolated bovine chondrocytes in an in vitro study by Metabolic Pharmaceuticals (patent applied [WO2013082667]). Its positive effects include promoting the repair of bone and cartilage in cases of OA.


It is well established that hGH is a lipolytic hormone (15), but the exact mechanisms used are still unclear. In this paper we present data that suggest that hGH and its lipolytic fragment (AOD9604) induce their chronic in vivo actions on lipolysis in part by modulating the expression of theβ 3-AR. Human GH has been shown to affect the in vivo expression and function of β-ARs in vivo in sheep (16). Data presented in this paper indicate that chronic administration of hGH influences expression of the β3-AR in adipose tissue in the ob/ob mouse. In brown adipose tissue (BAT), these compounds also increase expression of β3-AR expression in the lean C57BL/6J mouse. The increase in expression induced by chronic hGH or AOD9604 treatment correlated with the decrease in adipose tissue mass. We therefore hypothesize that treatment with either hGH or AOD9604 enhances β3-AR expression, which has been observed in murine 3T3-F442A and human SK-N-MC cells in vitro (11).

I was keen to try out CJC 1295 because my gym buddies had been using it for a while with fast and positive results. Though I was a bit nervous about injecting myself! Initially, I tried with the lowest dosage of CJC-1295 which was 10 mg for 10 weeks. It wasn’t look before I was seeing results. But, I have been advised to try out the CJC 1295 Ipamorelin combination, which I will do soon!
Up to the date of this paper, six clinical trials (three single dose studies and three multiple dose studies from which 2 were long-term studies) have been conducted with AOD9604. In none of the three acute dose-escalating studies (METAOD001 - METAOD003) were there reported any AEs related to the intake of AOD9604, withdrawals from the study caused by AOD9604 or SAEs reported. No treatment related differences were identified upon administration of AOD9604 by IV injection (25 µg - 400 µg/kg bodyweight) or orally (9 mg - 54 mg). Treatment with AOD9604 as a single dosage had no effect on physical examination, vital signs, laboratory parameters, ECG, blood glucose and IGF-1 levels, with results indistinguishable from placebo.

Resting plasma concentrations (mean±SEM) of mature ANP and Nt‐proANP were 14±2 pg/mL and 4±0.6 pg/mL at baseline. After gastric bypass surgery, the resting plasma concentrations rose to 24±5 pg/mL and 7±0.7 pg/mL for ANP and Nt‐proANP (increased by 23% and 43%), respectively (P=0.016 and 0.008). Absolute concentrations of ANP and Nt‐proANP were higher in individuals after bypass surgery at all acute time points during and after administration of intravenous saline (Figure 1A and 1B; P<0.001). A secondary analysis, adjusted for age, sex, and mean arterial pressure yielded similar results.

Paracetamol has long been considered very safe, without the risks of gastric injury associated with aspirin and NSAIDs. But there are distinct risks of liver injury, usually following overdose situations. In response many international regulatory authorities have taken steps to reduce the pack sizes of paracetamol, and to restrict release in some environments to pharmacies. In the USA, FDA has required prescription acetaminophen, when it is usually combined with an opioid, to reduce the dose per dose unit to 325 mg, but without reducing the maximal daily dose. No change of dosing in the USA has yet come for OTC acetaminophen. Use of paracetamol should be kept to a minimum in patients with underlying liver and renal disease. It can reduce the effects of lithium, ACE inhibitors, beta blockers and methotrexate. However, it remains one of the safest and most effective analgesic drugs, particularly in the elderly where the risks of gastric bleeding with NSAIDs are more common, and carries minimal side effects.
•If injecting just a GHRP or GHRH product on their own, avoid eating/drinking anything high in fat for 3 hours before your injection and anything high in carbohydrates for 2 hours before (i.e. always do your injection on an empty stomach), otherwise the amount of GH release they cause may be significantly blunted leading to poor results. •If injecting both a GHRP and GHRH together (e.g. 100mcg of both GHRP-6 and Modified GRF 1-29) studies have proven that their ability to release GH returns to full-strength as little as 1 hour (60 minutes) post-meal. This gives users greater flexibility with their meal timings, especially since consuming sufficient calories is so critical to building muscle. •Whether injecting GH peptides alone or along with others, always wait at least 20 minutes after your injection before consuming anything. Once at least 20 minutes has passed, consume a food/beverage high in protein and/or carbohydrates to stimulate an insulin spike (if you inject in the morning and around your workout, this meal/shake should be high protein and high carbohydrates, if you inject at night this consumption should be protein only as protein is sufficient enough to spike insulin, but without the negative impact on fat gain which carbohydrates can contribute to).
