Several epidemiologic studies have reported lower circulating natriuretic peptide concentrations in obese individuals.12, 14 However, these studies have been observational and confined to a single time point of measurement of natriuretic peptides. To our knowledge, only one previous study has examined the association of obesity with salt‐induced natriuretic peptide concentrations. Licata and colleagues found reduced, salt‐loaded plasma ANP concentrations in 9 obese individuals compared with 10 lean controls.21 They did not examine the influence of weight loss on the natriuretic peptide system. Thus, the present study is the first to provide serial, physiologic data from the same individuals over time.

In brown adipose tissue (BAT), these compounds also increase expression of β3-AR expression in the lean C57BL/6J mouse. The increase in expression induced by chronic hGH or AOD9604 treatment correlated with the decrease in adipose tissue mass. We therefore hypothesize that treatment with either hGH or AOD9604 enhances β3-AR expression, which has been observed in murine 3T3-F442A and human SK-N-MC cells in vitro (11).

Eligible study subjects were admitted after overnight fasting for an outpatient visit at the MGH CRC. In vitro studies by Metabolic Collaborators showed that AOD9604 enhances the differentiation of adipose mesenchymal stem cells into bone, promotes proteoglycan and collagen production in isolated bovine chondrocytes, and promotes differentiation of myoblasts into C2C12 cells. These effects induced by AOD9604 are similar to those required for the repair of bone, cartilage, and muscle, all of which are affected in OA. To the best of our knowledge, no study has compared the effects of GH and HA intra-articular injections on OA. A previous study showed the effects of intra-articular GH injection on articulophyseal cartilage regeneration in the knees of rabbits [9]. Our study showed that the groups that received AOD9604 or HA injection had better outcomes in terms of morphological and histolopathological findings, as well as a lowered duration of lameness than the group that received saline injections, although there are no significant differences between the two groups. In addition, our study revealed that the groups that received combined injections of AOD9604 and HA showed better outcomes than the groups that received AOD9604 or HA alone. The apparently synergistic effect of combined injections is thought to indicate that intra-articular injection of HA may have a chondrocyte-protective role, and the AOD9604 could help recapitulate the developmental cascades which regrows a segment of the articular cartilage in a joint. Our results are consistent with those of a previous study [8] that combined the injection of HA with recombinant human GH and found that the combination is more effective than HA injection alone.

Erosion of the articular cartilage, cleft, and cell cloning in the transitional and radial zones were noted in Group 3 (Figures 3-C,G,K). Softening of articular cartilage and surface irregularities were noted in Group 4 (Figures 3-D,H,L).
The objective of the 3rd study (METAOD003) was to assess the safety, tolerability and pharmacodynamics of single oral doses of AOD9604 in healthy, clinically obese males. 17 subjects (n = 15 completed the study), age 35 to 54 years, with a BMI ≥ 35 kg/m2 (range 35 to 56 kg/m2) subsequently received 3 increasing doses of AOD9604 (9, 27 and 54 mg) or placebo. Ultimately, it needs to be trialled in athletes. But that would never happen. As a substance with no therapeutic use, there is no need to ever test it to determine its effect on athletic performance. You don't need the same standard of evidence for drugs like EPO (synthetic replicas of hormones), where the primary biological effect studied in clinical trials of non-athletes is clearly and blatantly performance enhancing. With something like AOD9604, the waters are much murkier with respect to athletic performance.
AOD9604 is a new synthetic peptide fragment that comprises a modified 15 amino acid region of GH with a tyrosine component to help stabilize the molecule. Similar to GH, AOD9604 aids weight reduction in rodent models of obesity and was originally developed for the treatment of obesity in humans [11]. Design Mature New Zealand white rabbits (n=32) were randomly administered 2 mg collagenase type II twice in each knee joint. The black market uses for this peptide in muscle repair and treatment of obesity were also mentioned in the release, and it is clear through media reports on this scandal that Calzada gained traction from the profile boost (8).
Five of the submissions did not support the proposal while the sixth submission did. The former contend that potential risks of inadvertent use of caffeine in those at risk of an adverse event will be increased if selection of an analgesic is made without the assistance or intervention of a healthcare professional. There was also concern that the proposed exemption may result in an increase in liver damage due to excessive consumption of such a product. This was likely to result from people abusing these products as a source of stimulants.
As in the previous study there were no clinically relevant changes observed in safety laboratory parameters 24 hours following administration of AOD9604 or placebo. Similarly, there were no clinically relevant changes in vital signs (blood pressure, radial pulse rate and temperature) or ECGs recorded at any of the scheduled time points up to 24 hours post dose. There were no significant changes in glucose or IGF-1 levels following AOD9604 treatment compared with placebo.

We enrolled 15 obese individuals (mean BMI 45±5.4 kg/m2) undergoing gastric bypass surgery. Before and 6 months after surgery, subjects were admitted to the clinical research center and administered a large‐volume intravenous saline challenge. Echocardiography and serial blood sampling were performed. From the pre‐operative visit to 6 months after surgery, subjects had a mean BMI decrease of 27%. At the 6‐month visit, N‐terminal pro‐atrial NP (Nt‐proANP) levels were 40% higher before, during, and after the saline infusion, compared with levels measured at the same time points during the pre‐operative visit (P<0.001). The rise in Nt‐pro‐ANP induced by the saline infusion (≈50%) was similar both before and after surgery (saline, P<0.001; interaction, P=0.2). Similar results were obtained for BNP and Nt‐proBNP; resting concentrations increased by 50% and 31%, respectively, after gastric bypass surgery. The increase in NP concentrations after surgery was accompanied by significant decreases in mean arterial pressure (P=0.004) and heart rate (P<0.001), and an increase in mitral annular diastolic velocity (P=0.02).
The other submission commented on the consideration to place AOD-9604 in Appendix D. The submission supported listing in Schedule 4, but raised concerns that listing the substance in Appendix D would limit any future development work, including clinical trials that are currently being conducted on the substance. The submitter notes that there are currently 5 clinical trials notified to the TGA using this substance , with these approved clinical trials going ahead on the basis that the substance is safe for human use.
