I have been using sermorelin (bioidentical growth hormone releasing hormone) for 2 months now to help heal a nasty right quad tendon rupture suffered the end of December. I’m 52 years old with 7% bodyfat and am a lifetime strength trainer and former high level bike racer. 2 months ago, in spite of months of religious rehab, I couldn’t do a single right leg bench stepup. Yesterday I was doing 20lb DB’s for repeated sets of 15. I get complete blood panels every 6 months, and my last labs in May showed my IGF-1 levels off the reference range low. I get my next bloods in a couple of weeks. I was initially afraid to try this hormone due to the cancer implications, and I didn’t need it to be lean and fit, but I was desperate and for my injury recovery, and it has made a significant difference. The acute effect of AOD9604 and BRL37344 (aβ 3-AR agonist) on energy expenditure and substrate oxidation rates in WT and KO mice was also assessed. KO animals had lower energy expenditure, lower fat oxidation, and increased glucose oxidation, compared with the WT controls (data not shown). Injection of WT mice with a single dose of BRL37344 or AOD9604 increased energy expenditure and fat oxidation and decreased glucose oxidation. In the KO animals, BRL37344 failed to elicit any response in these metabolic parameters, clearly demonstrating that its effects are mediated exclusively through the β3-AR. AOD9604 did elicit a response in the KO mice, increasing fat oxidation and energy expenditure, although the response was not as great as in WT mice, suggesting that β3-ARs are not responsible for the acute biological response of AOD9604 on lipid metabolism. This is consistent with our previous findings in which AOD9604 was shown not to bind to the β3-AR (11). The size and duration of the metabolic responses to AOD9604 in the β3-AR KO animals was different from that observed in the control wild-type mice. The response was more rapid, shorter in duration, and greater in peak response. This may be because the KO animals are more acutely sensitive to lipolytic agents, a compensation for the ablation of the major lipolytic receptor.

It is well established that hGH is a lipolytic hormone (15), but the exact mechanisms used are still unclear. In this paper we present data that suggest that hGH and its lipolytic fragment (AOD9604) induce their chronic in vivo actions on lipolysis in part by modulating the expression of theβ 3-AR. Human GH has been shown to affect the in vivo expression and function of β-ARs in vivo in sheep (16). Data presented in this paper indicate that chronic administration of hGH influences expression of the β3-AR in adipose tissue in the ob/ob mouse. In brown adipose tissue (BAT), these compounds also increase expression of β3-AR expression in the lean C57BL/6J mouse. The increase in expression induced by chronic hGH or AOD9604 treatment correlated with the decrease in adipose tissue mass. We therefore hypothesize that treatment with either hGH or AOD9604 enhances β3-AR expression, which has been observed in murine 3T3-F442A and human SK-N-MC cells in vitro (11).

When AOD9604 or BRL37344 were administered to WT mice, an acute increase in fat oxidation and energy expenditure occurred, with an associated reduction in glucose oxidation (Fig. 6A). The effect plateaued 18 min following injection and remained stable for the duration of the experiment. The response to the two compounds was very similar, despite the fact we have previously shown that AOD9604 does not directly interact with the β3-AR as demonstrated by ligand binding studies (11). This clear separation of pathways was further confirmed in Fig. 6B in which AOD9604 clearly increases fat oxidation and energy expenditure inβ 3-KO mice, whereas BRL37344 does not. The KO mice neither decrease their glucose oxidation in response to AOD9604 nor show a prolonged increase in fat oxidation and energy expenditure in response to AOD9604.

The nature of the response to both hGH and AOD9604 is not clearly understood. We hypothesized that both molecules may influence the expression of the β3-adrenergic receptors (β3-ARs), the major lipolytic receptor in fat tissue. Both AOD9604 and hGH can increase β3-AR mRNA expression, as well as protein levels and function, in mouse and human cell lines in vitro (11). This response was investigated at the level of RNA and protein expression and function. The results for each mode of analysis were consistent in that both hGH and AOD9604 acted in a dose- and time-dependent manner to modulate the β3-AR response.

The duration (treatment and follow up) of the individual studies depended on the type of study (supplementary data). In June 2010, the National Drugs and Poisons Schedule Committee (NDPSC) considered the scheduling of paracetamol in combination with ibuprofen. Paracetamol preparations containing 500 mg or less of paracetamol as the only therapeutically active constituent (other than phenylephrine, effervescent agents or guaiphenesin) in packs of 25 or less were exempt from scheduling. However, when these preparations were combined with another therapeutically active ingredient they became Schedule 2. The NDPSC considered that the Schedule 2 entry remained appropriate, but noted the possibility that more robust evidence of additional risk could come to light through any application for product approval with the Therapeutic Goods Administration. The delegate confirmed the NDPSC's decision and the reasons for the decision in August 2010.
Time line of collagenase, saline, HA and AOD9604 injection. Collagenase (0.25 mg) was injected in right knee twice on day 1 and 4, respectively. Normal saline 0.6 ml (group 1), HA 6 mg (group 2), AOD9604 0.25 mg (group 3), and AOD9604 0.25mg with HA 6 mg (group 4) were injected in the right knee at 4 weeks after the first collagenase injection. All rabbits were euthanized by CO inhalation at 4 weeks after injection of 4 different solutions.
Press coverage and word of mouth has generated quite a bit of hype around AOD9604. Despite its popularity on the black market, there is no research into the safety and effect of injecting this peptide at high doses. Intriguingly, it is not currently in Calzada's interest to fight the black market production of AOD9604, even though it is a patented product. Calzada lack the funds to legitimately boost the reputation of this peptide through clinical trials, and there is a strong chance these would once again prove that AOD9604 has no effect. The good reputation this peptide currently enjoys is likely to provide strong leverage to its marketing as a food supplement in the United States.
From the standpoint of protein synthesis and muscle repair, IGF-1 injections have also been shown to enhance the anticatabolic effects of insulin and to increase the protein synthesis normally induced by growth hormone. This is because, like insulin, IGF-1 encourages amino acid uptake into muscle cells, stimulates peripheral tissue uptake of glucose (which lowers blood glucose levels), and suppresses liver glucose production. That last fact is important and is actually why IGF-1 is even being considered as a diabetes-prevention drug. Insulin resistance can cause the liver to produce excess glucose, which then causes even more insulin insensitivity and can eventually result in type II diabetes, and IGF-1 can decrease the need for this type excessive insulin release.
In the multiple dose and long term studies, AOD9604 was well tolerated over the entire dose range. In none of the studies did any drug-related withdrawals or drug-related serious AEs occur. No drug related clinically significant AEs, or changes of clinical significance in vital signs, safety laboratory tests or ECGs were detected during the studies. There were no observable trends in the incidence of AEs between the 0.25 mg, 0.5 mg, 1 mg, 9 mg and 27 mg AOD9604 and placebo treatment groups. The highest dose administration (54 mg), however, was associated with an increased incidence of GI-related AEs.
Ghrelin and GH secretagogues, including GH-releasing peptide (GHRP)-6, stimulate food intake and adiposity. Because insulin modulates the hypothalamic response to GH secretagogues and acts synergistically with ghrelin on lipogenesis in vitro, we analyzed whether insulin plays a role in the metabolic effects of GHRP-6 in vivo. Streptozotocin-induced diabetic rats received saline, GHRP-6, insulin, or insulin plus GHRP-6 once daily for 8 wk. Rats receiving saline suffered hyperglycemia, hyperphagia, polydipsia, and weight loss. Insulin, but not GHRP-6, improved these parameters (P < 0.001 for all), as well as the diabetes-induced increase in hypothalamic mRNA levels of neuropeptide Y and agouti-related peptide and decrease in proopiomelanocortin. Cocaine amphetamine-related transcript mRNA levels were also reduced in diabetic rats, with GHRP-6 inducing a further decrease (P < 0.03) and insulin an increase. Diabetic rats receiving insulin plus GHRP-6 gained more weight and had increased epididymal fat mass and serum leptin levels compared with all other groups (P < 0.001). In epididymal adipose tissue, diabetic rats injected with saline had smaller adipocytes (P < 0.001), decreased fatty acid synthase (FAS; P < 0.001), and glucose transporter-4 (P < 0.001) and increased hormone sensitive lipase (P < 0.001) and proliferator-activated receptor-gamma mRNA levels (P < 0.01). Insulin normalized these parameters to control values. GHRP-6 treatment increased FAS and glucose transporter-4 gene expression and potentiated insulin's effect on epididymal fat mass, adipocyte size (P < 0.001), FAS (P < 0.001), and glucose transporter-4 (P < 0.05). In conclusion, GHRP-6 and insulin exert an additive effect on weight gain and visceral fat mass accrual in diabetic rats, indicating that some of GHRP-6's metabolic effects depend on the insulin/glucose status.

I was using MT II while using the 1st vial and lost 14 lbs in 10 days. Had to fly to Pittsburgh for a couple days and started 2nd vial with no MT II and dropped 8 more. I weighed yesterday upon completion of the 2nd vial with shoes and clothes on. Was very pleased with the results. I did a cycle of T3 prior to the frag and didnt do near as well and was fighting muscle loss, felt like crap ect.. Will be purchasing more, I use Southern Research on the peptides.
A total of 97 AEs were reported by 17/17 subjects during this study. Most of them were of mild or moderate in intensity, with the exception of two SAEs, one of which (diarrhoea) was deemed “possibly related” to study treatment (54 mg AOD9604) and one (bronchial pneumonia) deemed to be “unrelated” to the study treatment (54 mg AOD9604). The most common adverse event reported was mild or moderate headache followed by events related to the digestive system, specifically diarrhea, flatulence, increased appetite and nausea. There was no observable trend between the AOD9604 groups or the placebo with respect to the incidence. The only event deemed definitely related to the treatment was taste perversion occurring 10 minutes following dose administration of the placebo.
Investigators at Monash University discovered that the fat-reducing effects of GH appear to be controlled by a small region near one end of the GH molecule. This region, which consists of amino acids 177-191, is less than 10% of the total size of the GH molecule and appears to have no effect on growth or insulin resistance. One submission was received, which did not support the delegate's interim decision, as available data support that the fixed dose paracetamol/caffeine combination product provides clinically meaningful efficacy over paracetamol alone; has an excellent safety profile; a very low risk of nephrotoxicity, toxicity in overdose, misuse, abuse or illicit use; and a highly favourable risk/benefit profile.
The study subjects were brought back to the MGH CRC 6 months after gastric bypass surgery and underwent an identical saline infusion protocol. Subjects were excluded from completing the second saline challenge protocol if they had developed complications of gastric bypass surgery including significant peri‐operative complications (myocardial infarction, persistent atrial fibrillation, sepsis, or gastrointestinal bleeding requiring blood transfusion >2 units).

There were no significant changes in IGF-1 values observed during 24 hours following any AOD9604 dose compared with placebo. No significant differences in glucose levels were observed following AOD9604 administration compared with placebo with the exception of one isolated time point (8% increase 12 hour post treatment in one subject receiving 54 mg AOD09604). There were no clinically significant observable trends in vital signs, physical examinations, abnormalities noted in the ECG measurements, or findings in the safety-related laboratory tests throughout the study.
I'm new to the forum and there is some great information here that I need to do more reading on. I've been taking Ipam and ModGrf 1-29 for at least 2yrs now. The first thing I noticed is that I have good quality sleep. I have difficulty sleeping and staying asleep. I take these peps 20min before I go to bed and get a good deep sleep for at least most of the night. I take them before doing my morning cardio, after my workout (afternoon) and before I go to bed. I've read if you take it before your morning cardio it releases more FFA to burn during your session. After the workout to aid in recovery. Before bed to aid in a deeper sleep. It is important when you take them. Dont eat before morning cardio...no carbs/sugary drinks...3hrs should have passed before taking another shot and do not eat before 15-20min after taking the peps. The reason (I've read) to wait these times is to take them while your insulin levels are low. High Insulin levels will minimize the pulsation of the GH. The phrase I've read is to try to keep "insulin quiet" to maximize the pulse of GH. Ipam can work by itself but if taken with ModGrf 1-29 it will magnify the GH Pulse. ModGrf is useless by itself. I've read that there is a saturation dose, so more is not better. 200mcg for each should do the job. Since synthetic GH shuts down natural production, these peps stimulate the pituitary to pulse more natural GH. To get the maximum effect of syn GH, you would also take the shot at the same time discussed above. Once again, this is what I've read and the protocol that I've followed. The results arent like AAS (nothing is!) but it is a good way to feel better since I cant take TRT. (trying to have kids) I also travel with the peps bc I value the sleep that I get from it.
Resting plasma concentrations of mature BNP and Nt‐proBNP were 14±3 pg/mL and 42±9 pg/mL before gastric bypass surgery and increased to 32±5 pg/mL and 107±20 pg/mL (increased by 50% and 31%), respectively (P=0.0009 and 0.0001) after the surgery. Circulating BNP and Nt‐proBNP concentrations during saline infusion were also higher after surgery compared with before surgery (Figures 2A and 2B; P<0.0001). Paracetamol has long been considered very safe, without the risks of gastric injury associated with aspirin and NSAIDs. But there are distinct risks of liver injury, usually following overdose situations. In response many international regulatory authorities have taken steps to reduce the pack sizes of paracetamol, and to restrict release in some environments to pharmacies. In the USA, FDA has required prescription acetaminophen, when it is usually combined with an opioid, to reduce the dose per dose unit to 325 mg, but without reducing the maximal daily dose.
