Phenylephrine is readily eliminated by sulphate conjugation in the intestinal wall, and oxidative deamination by monoamine oxidative glucuronidation in the liver. Monoamine oxidase (MAO) inhibitors can enhance the limited potential of phenylephrine for cardiac and pressor effects, by reducing metabolism. As a largely specific alpha adrenergic drug, with very weak beta agonism, there is little direct cardiac effect. However, in higher doses, there can be increases in both systolic and diastolic blood pressure and a reflex bradycardia. As an adrenergic agonist there is the potential to interact with other sympathomimetic drugs. In overdose phenylephrine can cause hypertension, headaches seizures tachycardia, and vomiting. There has been no evidence from carcinogenicity studies in rodents of any enhanced cancer risk over prolonged exposure.
In this study, plasma glycerol was also assayed as a measure of lipolytic rate. As shown in Fig. 5C, both AOD9604 and hGH increased glycerol levels following treatment in the WT mice, indicative of enhanced lipolysis. In the β3-KO mouse, however, no effect was observed with AOD9604. A significant increase in plasma glycerol from controls was observed following hGH treatment, but this was markedly less than that observed in the WT mouse. These data indicate the importance of β3-AR in the lipolytic response to AOD9604 in these animals and the necessity of the receptor for the chronic effectiveness of AOD9604 and hGH on fat reduction.

Streptozotocin-induced diabetic rats received saline, GHRP-6, insulin, or insulin plus GHRP-6 once daily for 8 wk. Rats receiving saline suffered hyperglycemia, hyperphagia, polydipsia, and weight loss. Insulin, but not GHRP-6, improved these parameters (P < 0.001 for all), as well as the diabetes-induced increase in hypothalamic mRNA levels of neuropeptide Y and agouti-related peptide and decrease in proopiomelanocortin. Cocaine amphetamine-related transcript mRNA levels were also reduced in diabetic rats, with GHRP-6 inducing a further decrease (P < 0.03) and insulin an increase. Diabetic rats receiving insulin plus GHRP-6 gained more weight and had increased epididymal fat mass and serum leptin levels compared with all other groups (P < 0.001). In epididymal adipose tissue, diabetic rats injected with saline had smaller adipocytes (P < 0.001), decreased fatty acid synthase (FAS; P < 0.001), and glucose transporter-4 (P < 0.001) and increased hormone sensitive lipase (P < 0.001) and proliferator-activated receptor-gamma mRNA levels (P < 0.01). Insulin normalized these parameters to control values. GHRP-6 treatment increased FAS and glucose transporter-4 gene expression and potentiated insulin's effect on epididymal fat mass, adipocyte size (P < 0.001), FAS (P < 0.001), and glucose transporter-4 (P < 0.05). In conclusion, GHRP-6 and insulin exert an additive effect on weight gain and visceral fat mass accrual in diabetic rats, indicating that some of GHRP-6's metabolic effects depend on the insulin/glucose status.

I have questions about combined therapy of CJC 1295 and Ipamorelin at the same time on a daily basis for both. The compounding pharmacies do not clearly state whether the CJC is with or without DAC. If it is the CJC with DAC, which sustains elevated GH and IGF-1 for several days, would taking it nightly in conjunction with the Ipramorelin, that is suggested to be taken TID but is being recommended only once at night, be over-stimulatory? If the CJC is without DAC, why take two pepetides simultaneously ,that have similar effects? I am just not clear why taking a daily dose of CJC with Ipamorelin as a single dose is better than taking the CJC with DAC twice per week alone or take the CJC with DAC for a while then switch to the Ipamorelin for a while?

Results: AOD 9604 had no effect on serum IGF-1 levels, which confirms the hypothesis that AOD 9604 does not act via IGF-1. Results of oral glucose tolerance test demonstrated that, in contrast with hGH, AOD 9604 has no negative effect on carbohydrate metabolism. Here we describe how hGH and AOD9604 can reduce body weight and body fat in obese mice following 14 d of chronic ip administration. These results correlate with increases in the level of expression ofβ 3-AR RNA, the major lipolytic receptor found in fat cells. Importantly, both hGH and AOD9604 are capable of increasing the repressed levels of β3-AR RNA in obese mice to levels comparable with those in lean mice. The importance ofβ 3-AR was verified when long-term treatment with hGH and AOD9604 in β3-AR knock-out mice failed to produce the change in body weight and increase in lipolysis that was observed in wild-type control mice. However, in an acute experiment, AOD9604 was capable of increasing energy expenditure and fat oxidation in theβ 3-AR knock-out mice. In conclusion, this study demonstrates that the lipolytic actions of both hGH and AOD9604 are not mediated directly through the β3-AR although both compounds increase β3-AR expression, which may subsequently contribute to enhanced lipolytic sensitivity.
