These findings suggest that the acute effects of AOD9604 are quite different from the chronic effects. Enhancedβ 3-AR expression appears to play a major role in the chronic effectiveness of the compound in terms of fat metabolism and weight loss. The acute effects observed in this study confirm that the β3-AR is not the sole mediator of this action. The increase in β3-AR expression in response to hGH and AOD9604 would permit enhanced lipolytic sensitivity. Identification of the components of the intracellular pathway(s) and effector(s) activated by AOD9604 are currently being investigated. The results presented in this paper suggest that the effectiveness of AOD9604 and hGH may partly rely on their ability to increase levels of β3-AR RNA expression in models of obesity in which the numbers of the lipolytic receptor are low. These unique properties may give AOD9604 an advantage over other lipolytic agonists such as adrenergic agents and hGH, which exhibit undesirable side effects when administered chronically (22).

Thirty-two rabbits were divided into 4 equal groups. Four different solutions, including saline, HA, AOD9604, and AOD9604 with HA, were injected in each group on a weekly basis for 4–7 weeks after the first collagenase injection. Group 1 received intra-articular saline injection (0.6 mL). Group 2 received intra-articular HA, (Hyruan-plus®; LG Life Science, Daejeon, Korea) injection (6 mg). The molecular weight of HA was measured at 3.0×106 Da, and it was prepared to a 10 mg/mL concentration. Group 3 received intra-articular AOD9604 (Metabolic pharmaceuticals, Melbourne, Australia) injection (0.25 mg per 0.6 mL). Group 4 received combined intra-articular AOD9604 (0.25 mg) and HA (6 mg) injections. All injections were administered by a physiatrist, using a commercially available ultrasound system with 3–12 MHz multi-frequency linear transducer (E-CUBE 15®; Alpionion Medical Systems, Seoul, Korea) under general anesthesia and under sterile conditions (Figure 1). No medication was administered after the injection. The rabbits were euthanized by CO inhalation 9 weeks after the first collagenase injection (Figure 2).

Prolonged use of intact hGH may result in negative effects on glucose metabolism, such as glucose intolerance and insulin resistance. Given that AOD9604 is a peptide fragment of the C-terminus of hGH, its effect on glucose control was monitored. OGTT testing was carried out on all patients during the screening phase as well as at various time points during the treatment period. No significant changes or obvious trends in the OGTT in any treatment group were observed, suggesting that AOD9604 supplementation does not deteriorate glucose control or induce insulin resistance. In contrast, treatment with AOD9604 seemed to have a positive effect in subjects with impaired glucose tolerance. In the 12 weeks treatment study, patients with impaired glucose tolerance supplemented with AOD9604 were less likely to develop diabetes during the study than subjects taking placebo. This hypothesis, however, has to be confirmed in a separate clinical trial. In contrast, in a recent clinical trial of hGH effects in obese women, hGH caused an increase in IGF-1 levels over a 6 month dosing period and decreased glucose tolerance in a subset of trial participants [29].

Whilst AOD9604 is not approved by the Australian TGA, it can be legally obtained on a doctor's prescription and dispensed by a compounding pharmacy (2). This is true of many experimental substances, but it does big favours for the reputation of AOD9604, giving the impression that, like other drugs issued by the medical profession, it is an efficacious and high quality product. These reasons, when considered together, give a powerful impression that peptides are highly effective – they wouldn't ban them for no reason, would they?
Resting plasma concentrations of mature BNP and Nt‐proBNP were 14±3 pg/mL and 42±9 pg/mL before gastric bypass surgery and increased to 32±5 pg/mL and 107±20 pg/mL (increased by 50% and 31%), respectively (P=0.0009 and 0.0001) after the surgery. Circulating BNP and Nt‐proBNP concentrations during saline infusion were also higher after surgery compared with before surgery (Figures 2A and 2B; P<0.0001). The saline infusion itself was not associated with an increase in BNP or Nt‐proBNP levels at either visit (P=0.65 and 0.60, respectively).

TGA evaluator concluded that the consistent absence of any clinically meaningful effects on blood pressure (BP) or heart rate (HR) in the applicant's bioavailability studies, and the absence of any ADR reports of BP, HR or other cardiovascular problems, indicate that "there is no valid reason for concern and no need to take any regulatory against the combination products currently in the ARTG and available in the Australian market", i.e. no demonstrated safety risk, and no evidence provided of efficacy of paracetamol 1000 mg / phenylephrine HCl 5 mg adult dose.

Ultimately, it needs to be trialled in athletes. But that would never happen. As a substance with no therapeutic use, there is no need to ever test it to determine its effect on athletic performance. You don't need the same standard of evidence for drugs like EPO (synthetic replicas of hormones), where the primary biological effect studied in clinical trials of non-athletes is clearly and blatantly performance enhancing. With something like AOD9604, the waters are much murkier with respect to athletic performance.
Mostly, these peptides are sold as lyophilized powder in 2mg containers. Bacteriostatic water should be mixed with the powder in order to reconstitute it. To make the dosage of 100mcg per injection, 2ml bacteriostatic water should be mixed into 2mg of lyophilized powder. This reconstituted mixture should be then injected inside the muscles or under the skin. The mixture should be kept under refrigeration at all times otherwise it will degenerate and will not be effective anymore.
The first was a double-blind, placebo-controlled, parallel group, multi-center study (5 Australian hospitals) (METAOD005). In this study 300 healthy obese males and females (BMI ≥ 35 kg/m2; Median BMI: 40 kg/m2, range: 35 to 67 kg/m2; 30 to 65 years old; 54% males and 46% females) were randomized to a 14-week period of daily oral dosing. The treatment period comprised a 2-week single-blind placebo run-in period followed by 12 weeks administration of either placebo or AOD9604 (1, 5, 10, 20 or 30 mg AOD9604 or placebo once daily; n =50 per group).

Other peptides amplify the body’s response to its own testosterone site-specifically within muscle and bone only. Steroids, on the other hand, produce their highly desired anabolic effects by acting on these receptors, but also come coupled with the less-sought-after side effects of acne, oily skin, hair loss, aggression, breast enlargement and testicular shrinkage.
