Injections of other compounds along with IGF-1 (which is a popular practice) can also cause serious health issues. The idea is that after an user administers a GHRP (like Ipamorelin) along with IGF-1, a selective pulse is then sent that stimulates the hypothalamus and pituitary to release even more growth hormone. Follow the same routine as shown above for "intermediate" persons. The expression of β3-AR RNA was assessed by RT by using radiolabeled primers and Southern blot analysis. Labeled bands were identified and semiquantitated by phosphorimaging. RT-PCR analysis demonstrated that ob/ob mice express lower amounts ofβ 3-AR RNA in their white (Fig. 3A) and brown (Fig. 3B) adipose tissues, compared with lean C57BL/6J mice, in agreement with others (14). Figure 3A shows the effect of saline, AOD9604, and hGH on β3-AR RNA expression in epididymal adipose tissue from both lean and obese mice, respectively. The level of β3-AR expression increased significantly in response to AOD9604 and hGH only in the obese mice, correlating with the significant decrease in epididymal adipose tissue weights seen in these mice. The same correlation was observed in brown adipose tissue, where increased expression of β3-AR was accompanied by a decrease in brown adipose tissue mass in both lean and obese mice (Fig. 3B).
Time line of collagenase, saline, HA and AOD9604 injection. Collagenase (0.25 mg) was injected in right knee twice on day 1 and 4, respectively. Normal saline 0.6 ml (group 1), HA 6 mg (group 2), AOD9604 0.25 mg (group 3), and AOD9604 0.25mg with HA 6 mg (group 4) were injected in the right knee at 4 weeks after the first collagenase injection. All rabbits were euthanized by CO inhalation at 4 weeks after injection of 4 different solutions.
In total 118 AEs were reported. No SAEs were reported. The most common adverse event reported by 16/23 subjects (69.6%) was mild or moderate headache. From all the reported AEs three events were reported of severe intensity (one in the 50 µg/kg AOD9604 group, 2 in the placebo group), with one of those events (a feeling of chest tightness) deemed possibly related to the AOD9604 treatment. Mild or moderate euphoria deemed possibly related to treatment, was reported by 5/23 subjects, during the periods when the AOD9604 was administered. There were no reports of euphoria during placebo administration. In total, there was no observable trend between the different treatment groups with respect to the incidence of certain AEs.
Finally, the hexadecapeptide AOD9604 did not induce allergenic reactions when consumed over 24 weeks. Blood of patients was analyzed for the presence of anti-AOD9604 antibody formation at various times and at the end of the studies (latest time point after 24 weeks). In none of the performed studies, at no time, were anti-AOD9604 antibodies detected in serum collected from any subjects in any treatment group.
